d-Cycloserine, a structural analog of d-alanine, is a broad-spectrum antibiotic produced by certain strains of Streptomyces orchidaceus or S. garphalus. d-Cycloserine is an FDA-approved medication classified as second-line drug for the treatment of tuberculosis already in human use since the 1950s. A few decades later, it was found that following penetration into the central nervous system (CNS), d-cycloserine has modulatory properties at the N-methyl-d-aspartate glutamate receptor (NMDAR) (Hood et al. 1989). Since NMDARs play a critical role in neuroplasticity, learning, and memory, current research focuses on the mechanisms of action and therapeutic potential of d-cycloserine in neuropsychiatric disorders including schizophrenia, cognitive dysfunctions, and affective and anxiety disorders.
