Abstract
The tidewater goby, Eucyclogobius newberryi, lives exclusively along the coast of California, primarily in high intertidal habitat seasonally isolated from the full effect of the tide. Rapid declines in the number of populations led to its listing as an endangered species in 1994. Here, we first reported the complete mitochondrial genome of this species, which is 16,526 bp in length, containing 13 protein-coding genes, two rRNA genes, 22 tRNA genes, a putative control region and one origin of replication on the light-strand (OL). The overall base composition is 25.6, 28.3, 27.8 and 18.3% for A, T, C and G, respectively. The 13 PCGs encode 3801 amino acids in total, twelve of which use the initiation codon ATG except COI use GTG. The most frequently used amino acid is leucine (16.24%), while cysteine acid (0.77%) is the least frequently used one. Besides, a phylogenetic tree based on the maximum likelihood method was constructed to explore the taxonomic status of E. newberryi, which strongly supported it had a closest relationship with the genus Gillichthys. These results would provide comprehensive phylogenetic relationship within Gobionellinae and further conservation strategies for this species.
