Summary
The problem was examined whether the genetical load of a human population due to the mutagenic action of cytostatic agents has reached an appreciable order of magnitude. The investigation was carried out in three steps:
-
1.
The number of persons treated with cytostatic drugs during their reproductive age (from 15 to 45) was estimated for the German Federal Republic and the year 1958, and the estimate was compared with the total population of the same age group. The number of children to be expected from treated persons under the assumption of age-specific normal reproduction was calculated. Both figures were found to be very low, indicating that even with normal reproduction of these persons, the additional load of mutations would be negligible.
-
2.
In a series of 663 patients treated with cytostatic drugs, reproduction after beginning of therapy was found to be reduced far below the age-specific level. This reduction was caused partially by biological factors, partially by socio-psychological influences.
-
3.
6 children were conceived after beginning of cytostatic treatment. None of them offered any hint for a possible genetic defect.
From the combined evidence derived from our three lines of argument, the conclusion seems to be warranted that at present, the genetic danger due to mutation induced by cytostatic treatment is very low, indeed. This, however, holds true only when cytostatic drugs are not used on a larger scale except for therapy of malignant diseases. Moreover, it does not mean that the genetic aspect can be neglected completely; experimental results in mammals point to possible genetic hazards for special families.
