An On-Chip Miniature Liquid Chromatography System: Design, Construction and Characterization

Abstract

Capillary liquid chromatography columns suitable for microchemical analysis have been fabricated on planar glass and silicon wafers, using standard microengineering techniques, including photolithography, wet chemical etching and anodic bonding. The columns are designed to allow the injection of sample solutions and elution solvents, and derivatisation of the inner walls. Columns modified with various stationary phases have been used to separate analytes, the detection of which is achieved by amperometric measurements.

The chromatography column forms part of an integrated system which, when completed, will comprise a miniature chemical measurement instrument, containing extraction and separation facilities. Fully portable, it will provide a cost-effective method of sample screening.
