Abstract

Problem 1) Maximum unknown, baseline increased, drug effects overadditively enhanced: the examination of dose response curves of functional synergism at different maxima reveals that, thereby, the combined effects (EA + b) never exceed the algebraic sum of the individual effects of A and B (EA + EB). Also, ED50 values will not be affected. Hence, larger combined effects and/or a decrease in ED50 indicates sequential potentiation (Pöch and Holzmann: J. Pharmacol. Meth. 4, 179, 1980) as observed, e. g., with isoprena-line in the presence of GTP on bovine coronary adenylate cyclase.
