Zusammenfassung
Frühere Immuntherapiestudien wurden meistens ohne begleitendes Immunmonitoring durchgeführt, so daß der Therapieeffekt nur anhand klinischer und bildgebender Diagnostik beurteilt werden konnte. Über die erzeugten immunologischen Effekte konnte jedoch nur spekuliert werden. Im Rahmen einer Phase I/II Vakzinstudie mit dem neuartigen, synthetisch hergestellten Impfstoff Baculovirus-GA733 sollte die Wertigkeit eines praktikablen Immunmonitorings mit Hilfe der Durchflußzytometrie untersucht werden. Das Tumor-assoziierte Antigen GA733, ein 40kD Glykoprotein, wird zu über 80% auf kolorektalen und Pankreaskarzinomen exprimiert [1, 2]. Die extrazelluläre Domäne des Antigens wurde in einem Baculovirus-System synthetisch hergestellt und mit Aluminiumhydroxid als Adjuvans intradermal injiziert.
