MRS Studies on Experimental Tumours Treated with TNF

Summary

Imaging-guided localized 31P MRS (4.7 tesla) were utilized to assess and monitor metabolic alterations induced by murine TNF-ß on solid tumours obtained by s.c. injection of Friend leukaemia cells in syngeneic mice. The cytokine induced significant reductions in the average ATP levels, associated with an increase of inorganic phosphate. These effects, detected at early times after cytokine administration are interpreted as due to induction of cell hypoxia associated with early blood vessel occlusion induced by the drug, before the occurrence of haemorrhagic necrosis. These results, together with previous in vitro MRS analyses of freshly dissected tissues demonstrated that the combination of MRI and 31P MRS methods provide useful means to monitor early bioenergetic alterations in experimental tumours in response to therapy and suggest possible extensions of these approaches to clinical investigations.
