Abstract
The greengram is a protein-rich legume is sensitivity to numerous biotic and abiotic stress leads to the huge loss to growers. Of the biotic stresses, Mungbean yellow mosaic virus (MYMV) is a severe threat for greengram production in country. The survey conducted in major greengram growing areas of Karnataka showed incidence of MYMV varied from 10.32 to 68.75 per cent. Greengram plants showing MYMV symptoms were collected from the different locations and confirmed with infection by PCR using virus specific primers. The complete genome of DNA-A and phylogeny showed that greengram isolates were closely clustered with MYMV infecting greengram, soybean and horsegram reported previously from different parts of India. The phylogenetic analysis of mtCOI gene from the B. tabaci was closely clustered with cryptic species belongs to the Asia-I group. The virus-vector interaction studies revealed that, minimum Acquisition Access Period (AAP) and Inoculation Access Period (IAP) for transmission of MYMV by whiteflies, Bemisia tabaci (Asia-I cryptic species) was 10 min. A single B. tabaci was sufficient to transmit the MYMV and seedlings susceptibility to MYMV was inversely related to its age. The study demonstrated that Asia-I cryptic species is an efficient vector of MYMV associated with greengram.
