Summary
The degree of sensitivity and of resolution of amino acids which is possible with TLC is much higher than with paper chromatography. This makes TLC on cellulose powder an ideal system for the analysis of protein hydrolysates and biological fluids, particularly where expense prohibits the use of the more elaborate automatic ion-exchange analysers. Using a new pair of solvents and a thin layer of cellulose,Heathcote &Jones were able to separate unambiguously some 23 amino acids chosen for their likelihood to appear in such fluids. The technique is capable of resolving difficult pairs of amino acids such as leucine/isoleucine, valine/methionine and glycine/serine in under 6 hours. Some of the practical considerations and applications of the technique will be discussed.

Le degré de sensibilité analytique et de résolution des amino acides qu'il est possible d'obtenir à l'aide de la CCM est beaucoup plus élevé qu'avec la chromatographie sur papier. Cela fait de la CCM sur poudre de cellulose un système idéal pour l'analyse des hydrolysats de protéines et des liquides biologiques, en particulier là où le coût élevé interdit l'emploi des analyseurs automatique par échange d'ions très développés. A l'aide d'un nouveau système de solvants et une couche mince de cellulose,Heathcote &Jones ont pu séparer d'une façon précise, quelques 23 amino acides sélectionnés selon la probabilité de leur apparition dans de tels liquides. Cette technique permet la résolution de couples difficiles d'amino acides tels que leucine/isoleucine, valine/methionine, et glycine/serine, en moins de 6 heures. Quelques considérations pratiques et quelques applications pratiques de cette technique seront discutées.
Zusammenfassung
Im Vergleich mit der Papierchromatographie sind der Bestimmungs-und Empfindlichkeitsgrad, die mit der DC erreicht werden, wesentlich höher. Die DC auf Zellulosepulver ist aus diesem Grunde ein ideales Analysenverfahren für Proteinhydrolysate und biologische Flüssigkeiten, insbesondere in Fällen, wo die hohen Kosten die Anwendung der sehr vervollkommneten, automatischen Analysatoren mittels Ionenaustausch verhindern. Mit Hilfe neuer Lösungsmittel und einer Dünnschicht von Zellulose konntenHeathcote &Jones mit Präzision einige 23 Aminosäuren trennen, die in solchen Flüssigkeiten vorkommen. Diese Technik erlaubt den Nachweis von schwierigen Aminosäure — Paaren wie z.B. Leucin/Isoleucin, Valin/Methionin und Glycin/Serin in weniger als 6 Stunden. Einige praktische Erwägungen und Anwendungen dieses Verfahrens werden diskutiert.
