Real-Time PCR and Antigen-Capture ELISA for the Diagnosis of Porcine Deltacoronavirus Infection

Abstract

Porcine deltacoronavirus (PDCoV) is a newly emerged swine enteric coronavirus that poses a major threat to the pig industry. Currently, there is no specific treatment or effective vaccines to prevent PDCoV infection. Effective diagnostic methods are essential for PDCoV infection. In this study, two quantitative PCR (qPCR) methods, TaqMan real-time quantitative PCR and SYBR Green I real-time PCR, were established for detecting viral nucleic acid based on the PDCoV M gene, and a double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) was developed for detecting viral antigen based on PDCoV N antibodies. All methods have high specificity and good reproducibility and can be used for clinical sample detection. These methods will be useful for the clinical diagnosis and epidemiological investigation of PDCoV.
