Abstract

A knowledge of intracellular activities is the prerequisite for an appraisal of concentration and electrochemical gradients established in living systems by their physical properties and metabolic activities. While the determination of cellular ionic concentrations appears at first to be relatively simple, it is in fact complicated by a number of factors:

  1. 1.

    Structural compartmentation of cells and tissues: This term involves not only the extracellular space in the analyzed biological material, but also intracellular structural compartmentation (subcellular components with distinct permeability barriers) as well as a possible non-homogenity of the analyzed cell population.

  2. 2.

    Chemical compartmentation, affecting the ionic activity in a given physical compartment, such as ‘binding’ of ions or availability of water as a solvent. While several approaches may be used for the determination of intracellular ionic concentrations or activities, the values obtained are mostly only approximations, and the term ‘apparent concentrations’ or ‘-activities’ appears to be justified.
