Abstract
In order to study the metabolic effects of the α-glucosidase inhibitor acarbose, eight normal subjects (four male and four female) and eight subjects with non-insulin- dependent diabetes mellitus (NIDDM) (six male and two female) were treated in a double-blind study with acarbose and placebo for 2 weeks each, in random order and with a 1-week washout period between. The normal subjects had a mean age (± SEM) of 46 ± 2 years and a relative body weight of 132 ± 4%. The NIDDM subjects had a mean age of 51 ± 4 years and a relative body weight of 118 ± 7%. The dose of acarbose was 200 mg before breakfast and 100 mg before lunch. At the end of each treatment period all subjects were given a standardized breakfast (625 kcal) at 0800 hours, normal mixed lunch (590 kcal) at 1130 hours, and a snack (75 kcal) at 1400 hours. Profiles of plasma glucose, serum insulin, serum C-peptide, and plasma GIP were obtained from 0800 to 1500 hours.
