Abstract
High-dose glucocorticosteroids are used successfully to prevent vomiting and nausea in patients undergoing cisplatin- and ifosfamide-containing chemotherapy, particularly in patients with testicular cancer. To rule out a possible effect of dexamethasone on the antitumor activity of cisplatin or ifosfamide, the proliferation of heterotransplanted human testicular tumors (H 12.1, H 12.7) growing subcutaneously in nude mice was studied during treatment with ifosfamide or cisplatin, given with or without dexamethasone.
In both cell lines a slight but statistically non significant retardation of tumor growth was observed with dexamethasone therapy. However, the antitumor activity of cisplatin or ifosfamide was not significantly influenced by concomitant administration of dexamethasone. From these data it is concluded that dexamethasone might slightly alter the growth of testicular tumors, having a minor therapeutic effect by itself, but that its administration as an antiemetic drug to patients treated with cisplatin or ifosfamide does not influence the antitumor activity of either drug.
Zusammenfassung
Hochdosierte Glukokortikosteroide werden mit Erfolg verwendet, um bei Patienten, die sich einer Chemotherapie mit Cisplatin bzw. Ifosfamid unterziehen, auftretende Übelkeit und Erbrechen zu vermeiden; diese Zytostatika sind essentielle Substanzen in der Therapie von Hodentumoren. Um einen möglicherweise bestehenden Effekt des Dexamethason auf die Antitumoraktivität von Cisplatin oder Ifosfamid zu untersuchen, wurde die Proliferation von subkutan auf Nacktmäuse heterotransplantierten humanen Hodentumoren der Zellinien H 12.1 und H 12.7 beobachtet, die mit Ifosfamid oder Cisplatin jeweils mit und ohne Dexamethasongabe behandelt wurden.
In beiden Zellinien wurde eine leichte, jedoch statistisch nicht signifikante Verzögerung des Tumorwachstums unter Therapie mit Dexamethason beobachtet. Bei gleichzeitiger Verabfolgung der Chemotherapeutika mit Dexamethason wurde die Antitumoraktivität von Cisplatin und Ifosfamid nicht signifikant beeinflußt. Aus diesem Ergebnis wird geschlossen, daß Dexamethason selbst das Wachstum von Hodentumoren geringfügig verändert, daß jedoch seine Verabreichung als Antiemetikum bei mit Cisplatin oder Ifosfamid behandelten Patienten mit Hodentumoren die Antitumoraktivität dieser beiden Substanzen nicht beeinflußt.
