Zusammenfassung
In den letzten Jahren sind zahlreiche inotrop wirkende Pharmaka zur Therapie der Herzinsuffizienz entwickelt worden [5]. Aus klinischer Sicht sind besonders diejenigen Substanzen von Interesse, welche für die Langzeittherapie geeignet sind und alternativ für oder zusätzlich zu einer Digitalismedikation verabreicht werden können. OPC-8212 - ein Chinolinonderivat (Abb. 1) - ist eine derartige inotrope Substanz. Nach tierexperimentellen Befunden führt OPC-8212 zu einer Hemmung der Phosphodiesterase [6, 7]. Möglicherweise tragen jedoch noch andere Effekte zur Substanzwirkung bei [6, 8]. Hinsichtlich seiner pharmakokinetischen Eigenschaften unterscheidet sich OPC-8212 jedoch durch seine lange Eliminationshalbwertzeit deutlich von anderen neuen inotrop wirkenden Pharmaka.
