Thromboxane and Prostacyclin Production by Irradiated and Perfused Rat Kidney

Abstract

The effect of ionizing radiation on renal prostaglandin synthesis was investigated in rats subjected to a single radiation fraction of 15 Gy administered to the left kidney. Seven days later, irradiated kidneys and control kidneys from sham-irradiated animals were excised and perfused with a buffer to allow the evaluation of prostaglandin synthesis by the tubular and the capillary systems. Effluent fluids collected from the renal vein and ureter were assayed by radioimmunoassay for thromboxane A2 (TXB2) and prostacyclin (6-keto-PGF). Angiotensin II stimulation was used to determine the capacity of the kidneys to synthesize the above prostaglandins. Morphological changes observed 1 week following irradiation were minimal and consisted of interstitial edema; however, changes in prostaglandin synthesis were profound. A significant increase in the basal and stimulated releases of thromboxane A2 was observed in the ureteral and venous effluents of irradiated kidneys whereas the basal and stimulated releases of prostacyclin were increased only in the ureteral effluents. These changes may be involved in the pathogenesis of the postradiation capillary and tubular injuries.
