First report of Papaya ringspot virus-watermelon strain on melon (Cucumis melo L.) in Bali, Indonesia

Abstract

Symptoms of mosaic, with puckering and blistering on leaves, were found in cultivated melon in 2020 in Bali, Indonesia. RT-PCR, a universal primer of CI gene of Potyvirus, successfully amplified a DNA sized ± 683 bp. The homologies of nucleotide (nt) and amino acid (aa) sequences of PRSV-W Bali isolate to those isolates from other countries, was 94.1–98.8% nt and 94.6–99.1% aa. The cylindrical inclusion nucleotide and amino acid sequences of three isolates, from Bali, had the highest identity with that of PRSV-W isolate cucurbit in East Timor. PRSV-W Bali was in the same cluster with PRSV-W from East Timor and Australia. By mechanical inoculation and aphid transmission, the virus was able to infect systemically melon plants. This is the first report of PRSV-W infection on melon in Indonesia.
